Chickens were immunized with the 6X-Histidine peptide conjugated to keyhole limpet hemocyanin protein and emulsified in Freund’s adjuvants. After multiple injections, eggs were collected from the hens, IgY fractions were prepared from the yolks, and then affinity-purified antibodies were prepared using 6X-Histidine peptide conjugated to an agarose matrix. To make up the final mixture, the IgY fraction was spiked with affinity purified antibody, mixed with glycerol and sodium azide, and filter-sterilized.
Volume: 100 µL (HIS-W-1010) or 400 µL (HIS-W-1040)
Concentration: 1 mg/mL
Host Species: Chicken
Buffer: Sodium phosphate-buffered (10 mM) isotonic (0.9%, w/v) saline ("PBS," pH 7.2) (50% by volume) with glycerol (50% by volume) as an anti-freezing agent and sodium azide (0.02%, w/v) as a preservative.
Validation and Application Notes
IHC Dilution Range: 1:5000-1:10000
Store at -20°C in the dark. Under these conditions, these antibodies should have a shelf life of at least 12 months (provided they remain sterile). Do not freeze these antibodies at –80°C unless you want to store them for longer periods of time. Note, however, that each time an antibody preparation is frozen, about half its binding activity is lost.
